The Collector's Edition at $249.99 is primarily for die-hard collectors who want the 12-inch statue and steelbook — the digital content is identical to Deluxe.

No, the digital content is identical between Deluxe ($99.99) and Collector's Edition ($249.99). The $150 premium is entirely for physical items: a 12-inch statue, steelbook case, cloth map, playing cards, keychain, and art book.
5 days of early access starts November 14. If you have the time to play, it is worth it for avoiding launch-day server issues and being among the first to explore Vice City. If you work weekdays, you may see limited benefit.
Rockstar has not announced whether the Leonida Story Pack will be available as standalone DLC. Historically, GTA IV's exclusive content never became available separately. Assume it is Deluxe-only permanently.
